Osteogenic molecules for clinical applications: improving the BMP-collagen system Biol. Res.
Arrabal,Pilar M; Visser,Rick; Santos-Ruiz,Leonor; Becerra,José; Cifuentes,Manuel.
Among the osteogenic growth factors used for bone tissue engineering, bone morphogenetic proteins (BMPs) are the most extensively studied for use in orthopaedic surgery. BMP-2 and BMP-7 have been widely investigated for developing therapeutic strategies and are the only two approved for use in several clinical applications. Due to the chemical and biological characteristics of these molecules, their authorised uses are always in combination with a carrier based on collagen type I. Although the use of these growth factors is considered safe in the short term, the very high doses needed to obtain significant osteoinduction make these treatments expensive and their long-term safety uncertain, since they are highly pleiotropic and have the capacity to induce...

An ultrasound and histomorphological analysis of experimental liver cirrhosis in rats BJMBR
Dias,J.V.; Paredes,B.D.; Mesquita,L.F.Q.; Carvalho,A.B.; Kozlowski,E.O.; Lessa,A.S.; Takiya,C.M.; Resende,C.M.C.; Coelho,H.S.M.; Campos-de-Carvalho,A.C.; Rezende,G.F.M.; Goldenberg,R.C.S..
We investigated whether liver injury by dual exposure to ethanol and carbon tetrachloride (EtOH + CCl4) for 15 weeks would persist after hepatotoxic agents were removed (EtOH + CCl4/8wR). After 15 weeks of hepatic injury with ethanol (5.5%, m/v) and carbon tetrachloride (0.05, mL/kg, ip), 5 of 11 female Wistar rats were sacrificed. The other 6 rats were maintained for an additional 8 weeks without hepatotoxic agents. Ultrasonography showed increased liver echogenicity and dilation of portal vein caliber in both groups (EtOH + CCl4: 0.22 ± 0.01 cm, P < 0.001; EtOH + CCl4/8wR: 0.21 ± 0.02 cm, P < 0.01) vs control (0.16 ± 0.02 cm). Histopathology showed regenerative nodules in both experimental groups. Histomorphometry revealed increased fibrosis...

Review of the Y chromosome and hypertension BJMBR
Ely,D.; Turner,M.; Milsted,A..
The Y chromosome from spontaneously hypertensive rats (SHR) has a locus that raises blood pressure 20-25 mmHg. Associated with the SHR Y chromosome effect is a 4-week earlier pubertal rise of testosterone and dependence upon the androgen receptor for the full blood pressure effect. Several indices of enhanced sympathetic nervous system (SNS) activity are also associated with the SHR Y chromosome. Blockade of SNS outflow reduced the blood pressure effect. Salt sensitivity was increased by the Y chromosome as was salt appetite which was SNS dependent. A strong correlation (r = 0.57, P<0.001) was demonstrable between plasma testosterone and angiotensin II. Coronary collagen increased with blood pressure and the presence of the SHR Y chromosome. A...

Corticosteroids in acute respiratory distress syndrome BJMBR
Fernandes,A.B.S.; Zin,W.A.; Rocco,P.R.M..
Improving the course and outcome of patients with acute respiratory distress syndrome presents a challenge. By understanding the immune status of a patient, physicians can consider manipulating proinflammatory systems more rationally. In this context, corticosteroids could be a therapeutic tool in the armamentarium against acute respiratory distress syndrome. Corticosteroid therapy has been studied in three situations: prevention in high-risk patients, early treatment with high-dose, short-course therapy, and prolonged therapy in unresolving cases. There are differences between the corticosteroid trials of the past and recent trials: today, treatment starts 2-10 days after disease onset in patients that failed to improve; in the past, the corticosteroid...

Contribution Towards the Anatomy of the Esophageal Hiatus and its Relationship with the Presence of Bundles of Collagen Fibers in its Margins International Journal of Morphology
Filho,José Jeová de Oliveira; Filho,Benedito Herani; Reis,Francisco Prado; Feitosa,Vera Lúcia Correa; Aragão,José Aderval.
The aim was to detect the presence of bundles of collagen fibers in the margins of the esophageal hiatus and establish if there is any relationship of these bundles with the anatomy of the hiatus. Ten adult male cadavers, with no gross anatomical alteration caused by trauma, surgery or disease, upon the esophageal hiatus were used in the study. A piece of anatomical structure comprising the diaphragm with the esophageal hiatus and adjacent tissues was removed, dissected and 10% formol embedding. With the aid of a digital caliper, measurements of the perimeter of the esophageal hiatus were done both in the abdominal and thoracic sides. For the structural study each margin was divided in six sections. Staining techniques of Masson and Picrosirius-hematoxilin...

Modulation of extracellular matrix by nutritional hepatotrophic factors in thioacetamide-induced liver cirrhosis in the rat BJMBR
Guerra,R.R.; Trotta,M.R.; Parra,O.M.; Avanzo,J.L.; Bateman,A.; Aloia,T.P.A.; Dagli,M.L.Z.; Hernandez-Blazquez,F.J..
Nutritional substances associated to some hormones enhance liver regeneration when injected intraperitoneally, being denominated hepatotrophic factors (HF). Here we verified if a solution of HF (glucose, vitamins, salts, amino acids, glucagon, insulin, and triiodothyronine) can revert liver cirrhosis and how some extracellular matrices are affected. Cirrhosis was induced for 14 weeks in 45 female Wistar rats (200 mg) by intraperitoneal injections of thioacetamide (200 mg/kg). Twenty-five rats received intraperitoneal HF twice a day for 10 days (40 mL·kg-1·day-1) and 20 rats received physiological saline. Fifteen rats were used as control. The HF applied to cirrhotic rats significantly: a) reduced the relative mRNA expression of the genes: Col-α1 (-53%),...

Biaxial testing of canine annulus fibrosus tissue under changing salt concentrations Anais da ABC (AABC)
Huyghe,Jacques M..
The in vivo mechanics of the annulus fibrosus of the intervertebral disc is one of biaxial rather than uniaxial loading. The material properties of the annulus are intimately linked to the osmolarity in the tissue. This paper presents biaxial relaxation experiments of canine annulus fibrosus tissue under stepwise changes of external salt concentration. The force tracings show that stresses are strongly dependent on time, salt concentration and orientation. The force tracing signature of are sponse to a change instrain, is one of a jumpin stress that relaxes partly as the new strain is maintained. The force tracing signature of a stepwise change in salt concentration is a progressive monotonous change in stress towards a new equilibrium value. Although the...

Collagen arrangement in hepatic granuloma in mice infected with Schistosoma mansoni: dependence on fiber radiation centers BJMBR
Lenzi,H.L.; Kimmel,E.; Schechtman,H.; Pelajo-Machado,M.; Vale,B.S.; Panasco,M.S.; Lenzi,J.A..
The collagen structure of isolated and in situ liver granuloma from Swiss Webster mice infected with Schistosoma mansoni was sequentially and three-dimensionally analyzed during different times of infection (early acute, acute, transitional acute-chronic, and chronic phases) by laser scanning confocal microscopy and electron scanning variable vacuum microscopy. The initial granuloma structure is characterized by vascular collagen residues and by anchorage points (or fiber radiation centers), from where collagenous fibers are angularly shed and self-assembled. During the exudative-productive stage, the self-assembly of these fibers minimizes energy and mass through continuous tension and focal compression. The curvature or angles between collagen fibers...

Placental osseous metaplasia associated with abortion in a mare: case report Arq. Bras. Med. Vet. Zootec.
Lima,P.A.; Eckstein,C.; Ecco,R.; Santos,R.L..
ABSTRACT The aim of this report is to describe a case of marked osseous metaplasia in the placenta associated with abortion in a mare. A 19-year-old Campolina mare with a history of multiple parturitions and absence of reproductive failure, aborted a fetus with approximately six months of gestation. There were no gross changes in the fetus and the placenta. Microscopically, there was multifocal replacement of fetal connective mesenchyme of the chorionic villi with well differentiated and partially mineralized osseous tissue (confirmed by von Kossa staining). To further characterize the collagenous component of the lesion, sections of the placenta were stained with Masson’s trichrome and Picrosirius red. Osseous tissue deposition had abundance of type I...

Adrenomedullin and adrenotensin regulate collagen synthesis and proliferation in pulmonary arterial smooth muscle cells BJMBR
Li,W.; Kong,Q.Y.; Zhao,C.F.; Zhao,F.; Li,F.H.; Xia,W.; Wang,R.; Hu,Y.M.; Hua,M..
To understand the pathophysiological mechanisms of pulmonary arterial smooth muscle cell (PASMC) proliferation and extracellular-matrix accumulation in the development of pulmonary hypertension and remodeling, this study determined the effects of different doses of adrenomedullin (ADM) and adrenotensin (ADT) on PASMC proliferation and collagen synthesis. The objective was to investigate whether extracellular signal-regulated kinase (ERK1/2) signaling was involved in ADM- and ADT-stimulated proliferation of PASMCs in 4-week-old male Wistar rats (body weight: 100-150 g, n=10). The proliferation of PASMCs was examined by 5-bromo-2-deoxyuridine incorporation. A cell growth curve was generated by the Cell Counting Kit-8 method. Expression of collagen I,...

Histochemical study of fibrillar proteins of the extracellular matrix in benign and malignant mammary neoplasms in dogs Braz. J. Vet. Res. Anim. Sci.
Martins,Ana Maria Cristina Rabello Pinto da Fonseca; Tamaso,Elia; Guerra,José Luiz.
The aim of the present study was to study some of the proteins that form the extracellular matrix of 54 benign and malignant mammary neoplasms in dogs, using histochemical methods: Picrosirius and polarization microscopy for collagenous fibers, Gordon -Sweats's method for reticular fibers and Weigert's fucsine-resorcine method for elastic fibers. A large variability in quantity, distribution and characteristics of the matrix components was observed in the different types of neoplasms. Collagen type I, III and elements of the elastic system had different distribution in benign and malignant neoplasms. The simple Picrossirius method and under polarization enabled visualization of collagen as thick fibers irregularly distributed in the stroma of carcinomas...

Lean vs. Obese Mice: The Ventral Prostate Revisited International Journal of Morphology
Martins,Fabiane Ferreira; del Sol,Mariano; Aguila,Marcia Barbosa; Mandarim-de-Lacerda,Carlos Alberto.
Obese mice (C57BL/6J-ob/ob) do not express leptin and develops hyperphagia, decreased energy expenditure, obesity, hyperglycemia, hyperinsulinemia, hypothermia, and infertility. Obesity causes reproductive dysfunction with negative impacts on prostatic structure and fertility. We aimed to compare the structure and molecular aspects of the ventral prostate between of lean and obese (ob/ob) mice. Three months old male lean and obese mice had their prostates dissected and prepared for light microscopy and immunofluorescence. In comparison to the lean mouse, the obese mouse showed a substantial structural modification in the ventral prostate starting with an atrophy of the prostate ventral lobe. Histologically, the acini showed a reduction in size, and in the...

Supraorganized collagen enhances Schwann cell reactivity and organization in vitro BJMBR
Maturana,L.G.; Zanon,R.G.; Pierucci,A.; Vidal,B.C.; Oliveira,A.L.R..
We investigated the reactivity and expression of basal lamina collagen by Schwann cells (SCs) cultivated on a supraorganized bovine-derived collagen substrate. SC cultures were obtained from sciatic nerves of neonatal Sprague-Dawley rats and seeded on 24-well culture plates containing collagen substrate. The homogeneity of the cultures was evaluated with an SC marker antibody (anti-S-100). After 1 week, the cultures were fixed and processed for immunocytochemistry by using antibodies against type IV collagen, S-100 and p75NTR (pan neurotrophin receptor) and for scanning electron microscopy (SEM). Positive labeling with antibodies to the cited molecules was observed, indicating that the collagen substrate stimulates SC alignment and adhesion (collagen IV...

Soft Collagen-Gelatine Sponges by Convection Drying BABT
Meyer,Michael; Trommer,Kristin.
This study showed that thermally labile fibrillar collagen could be processed continuously in combination with gelatine as foaming additive by convection drying. The procedure led to stable sponges with similar structural and physical properties as found for freeze-dried collagen samples. The fibrillar collagen remained native, while gelatine acted as foaming additive. The absorbency of the sponges was improved by opening the surface with abrasives. A use as medical device with hemostyptic properties would be possible.

Collagens and proteoglycans of the corneal extracellular matrix BJMBR
Michelacci,Y.M..
The cornea is a curved and transparent structure that provides the initial focusing of a light image into the eye. It consists of a central stroma that constitutes 90% of the corneal depth, covered anteriorly with epithelium and posteriorly with endothelium. Its transparency is the result of the regular spacing of collagen fibers with remarkably uniform diameter and interfibrillar space. Corneal collagen is composed of heterotypic fibrils consisting of type I and type V collagen molecules. The cornea also contains unusually high amounts of type VI collagen, which form microfibrillar structures, FACIT collagens (XII and XIV), and other nonfibrillar collagens (XIII and XVIII). FACIT collagens and other molecules, such as leucine-rich repeat proteoglycans,...

Polysaccharides of marine origin are gaining interest as biomaterial components. Bacteria derived from deep-sea hydrothermal vents can produce sulfated exopolysaccharides (EPS), which can influence cell behavior. The use of such polysaccharides as components of organic, collagen fibril-based coatings on biomaterial surfaces remains unexplored. In this study, collagen fibril coatings enriched with HE800 and GY785 EPS derivatives were deposited on titanium alloy (Ti6Al4V) scaffolds produced by rapid prototyping and subjected to physicochemical and cell biological characterization. Coatings were formed by a self-assembly process whereby polysaccharides were added to acidic collagen molecule solution, followed by neutralization to induced self-assembly of...
